Electroporation was used to introduce the hygromycin phosphotransferase reporter gene into creeping bentgrass (Agrostis palustris [A. stolonifera var. palustris]) protoplasts isolated from embryogenic suspension cells. After the subsequent selections with 50 mg hygromycin/litre, several plantlets were regenerated from hygromycin resistant calluses. PCR and Southern hybridization analysis of randomly chosen plants indicated that the transgene was integrated into their genomes and no false positiveswere found. The results suggested that hygromycin phosphotransferase gene was effective selectable marker gene for the selection of transformed cells. Overall frequency for hygromycin resistant colony formation in this report was 1-3 X 10 -6 based on thenumber of treated protoplasts.
