Plum pox potyvirus (PPV) was previously the only Potyvirus known to infect stone fruits. Recently, a new Potyvirus, asian prunus latent potyvirus (APLV), was reported to infect Prunus spp. The virus reacts positively in ELISA with PPV antiserum fromFrance, but negatively with PPV DNA primers specific for the 3' non-coding region (NCR) of PPV in RT-PCR. Biological and serological testing suggested the possible presence of PPV in sweet cherry (Prunus avium cv. Ferrovia) from southern Italy. To determine whether the virus in sweet cherry was PPV or APLV, the 3' NCR DNA primers specific for PPV were used to assay for its presence in extracts of sweet cherry leaves determined to be positive by ELISA. Uninfected cherry tissue and PPV-infected plum tissue were used as controls. RT-PCR-amplified DNA products from infected sweet cherry and plum tissue were of the expected size, 220 bp. No amplified products were obtained from healthy controls. The 3' NCR product from PPV-infected sweet cherry was isolated from 5% polyacrylamide gels and directly cloned using the TA cloning system. cRNA probes generated using T7 RNA polymerase were PPV specific. Nucleotide sequencing revealed the clones to be a portion of PPV genome. The results confirmed that the virusin sweet cherry in Italy is an isolate of PPV.
