In 2019, the European Medicines Agency (EMA) recommended adding a warning about a teratogenic risk to the summary of product characteristics (SPC) for modafinil-containing products (1,2).This decision followed the collection of 51 reports in the European pharmacovigilance database and the results of a United States registry study (2,3). Analysis of the registry in 2018 showed a rate of major congenital malformations of 15% (including heart defects, hypospadias and cleft palate) compared to 3% in the general population (4).
