摘要

The neutrophil-specific antigen NA is biallelic, composed of NA1 and NA2.NA antigens are often targets of antibodies which cause immune neutropenia. The phenotype of the NA system has been determined by the reaction of specific antibodies. We developed a new NAl-specific monoclonal antibody, TAG1. The reactivity of TAG1 to neutrophils was identical to that of MG38, which has been previously used to detect NA1 antigen. TAG1 did not react with any other blood cells or with cell lines derived from hematological malignancies, suggesting specificity for mature neutrophils. Preincubation of NAl/NAl neutrophils with MG38 remarkably abolished the binding of TAG1. Further, the reactivity of TAG1 to solubilized antigen from NA1 neutrophils was significantly lower than that of MG38 by the mixed passive hemagglutina-tion method. These results suggest that TAG1 specificically recognizes the NA1 antigen through different epitopes to those detected by MG38.
