We studied the occurance of DNA damage in leucocytes and separated mononuclear cells (MN) of 10 subjectes after a half marathon run (HM) using the single cell gelelectropho-resis (Comet-) assay. Additionally the cyto-plasmic expression of the antioxidant stress-protein heme oxygenase-1 (HO-1) in lympho-(L), mono- (M) and granulocytes (G) was analysed by flow cytometry. Total leucocytes exhibited a greater frequency of DNA damaged cells (33%) 24 h after HM compared to separated mononuclear cells (15%), indicating a greater extent of DNA effects in G. Single MN showed a pronounced extent of DNA damage and the frequency of DNA effects in MN 24 h after HM was related to changes in plasma TNFa directly after exercise. Cytoplasmic HO-1 rose in L, G and M, but G and M exhibited a more pronounced expression 0, 3 and 24 h after HM. Differences in DNA damage and the expression of HO-1 between the leucocyte populations after HM let us suggest, that M and G are critical target cells susceptible to exercise induced oxidative stress.
