Outcompeting neighbors for intestinal cancer initiation

Χntercellular communication is an important feature of multicellular organisms. However, these open avenues of communication can also be used by one set of cells to the detriment of their neighbors, which occurs during the process of cell competition, in which viable cells are eliminated by neighboring, fitter cells because of interactions that take place between them. Χuch of our knowledge on cell competition comes from studies in Drosophila, and the role of this process during mammalian tumor initiation has remained less explored. Χhree papers published this year in Nature now fill this gap by showing that cell competition has a key role in the initiation of intestinal cancer, by facilitating increased colonization of intestinal crypts by intestinal stem cells (ISCs) that contain oncogenic mutations. Notably, cell competition in this setting did not result in apoptosis of the outcompeted wild-type ISCs, but instead resulted in a significant growth disadvantage in the form of attenuation of their stemness properties. This was found to be caused by increased expression and secretion of WNT and BMP pathway ligands by the mutant cells, paving the way for them to thrive and initiate tumors by colonizing the intestinal crypts.
