Drought stress has a critical role to reduce plant growth and also alter composition and quantity of secondary metabolites. Secondary metabolites generally have been used in medicinal and food industry. Moreover, application of bio-chemical compounds such as chitosan has been reported to enhance tolerance of plants under abiotic and biotic stress. Therefore, this study was carried out to illuminate how chitosan functions in response to drought stress and its effect on secondary metabolism in Carum copticum L. (ajwain) under in vitro condition by calculating some physiological parameters and secondary metabolites content. Seedlings and callus of ajwain were cultured in MS medium with 0, 3 and 6% mannitol and different levels of chitosan (0, 10 and 20 mg/L). After 4 weeks, the results indicated that drought stress negatively affected germination, root and shoot length and fresh weight of seedlings. In response to drought stress, the content of anthocyanin, phenolic components, root carbohydrates, proline, and activity of some antioxidants enzymes significantly increased in ajwain. Additionally, chitosan improved total chlorophyll, carotenoids and subsequently carbohydrate content increased in root and shoot of ajwain under drought stress. Chitosan treatments increased the content of p-cymene, and thymol in calli and seedlings of ajwain, while the amount of gamma-terpinene decreased in both stage of growth. It seems that chitosan might improve tolerance of ajwain to drought stress by altering plant metabolism, and scavenging properties.
