Actinomycetes derived from unfamiliar surroundings are considered as a promising supply fornovel bioactive compounds comprising a broad range of biological actions. The current study was carried outto find out the bioactive adjacent to resistant pathogenic strains. During the study, 46 strains of actinomyceteswere isolated from the soil sediment sample collected from Manasbal Lake of Kashmir. During the primaryscreening, 14 actinomycete isolates showed an inhibition zone against MTCC pathogenic cultures, includingclinical isolates of Streptomyces aureus. Out of 14 isolates, 8 showed a wide range of activity. One isolate A41was found to be propitious for its wide spectrum of antibacterial action. Phylogenetic studies of A41 strainshowed maximum resemblance with Streptomyces exfoliates MMA1033. The study was extended to standardizethe fermentation conditions to enhance bioactive metabolite production. For A41, actinomycetes isolationagar (AIA) was confirmed as the best growth medium for maximum antibiotic production on the 16th day atpH 7, with an inoculum size of 2.5% (w/v) at 28℃. Peptone 1% (w/v) and sucrose 1% (w/v) were optimizedas the best nitrogen and carbon sources respectively for the highest antimicrobial production. The isolateswere not found to be halotolerant, exhibited the highest antibiotic production at 1.5% (w/v) of NaCl. Theadvance exploration could provide us extremely resourceful antibacterial agents in opposition to the clinicalimperative human pathogens. To the best of our knowledge, this is the first report on an assorted assembly ofActinomycetes from freshwater lakes of Kashmir having wide spectrum antimicrobial action.
