Sugarcane (Saccharum offidnarum L.), an important plant that is widely grown in tropical and subtropical regions, can be severely damaged by insect pests. Among these pests, Lepidiota signata Fabricius, a species of the order Coleoptera and the family Scarabaeidae, is the most devastating one for sugarcane. To prevent sugarcane crop from such attack, one of the best strategically optimal methods that can be used is the application of Bacillus thuringiensis (Bt) transgenic technology to generate new sugarcane varieties resistant to insects in a short time. In this study, the Bt insecticidal gene cry8Db and the selection marker gene hpt, were promoted by Ubiquitin promoter and combined into pCAMBIA1300 vector. The plant expression vector containing cry8Db gene was introduced into embryogenic callus of sugarcane variety KK3 via Agrobacterium tumefaciens. Transgenic sugarcane plants were selected by culture medium supplemented with hygromycine 50 mg/L. Eight survived sugarcane plants were tested by PCR for the presence of cry8Db gene with five of these being positive. The results of ELISA and Western blot analysis showed that five sugarcane lines possessed Cry8Db protein with the concentrations of Cry8Db protein ranging from 0.69 ng/g to 5.21 ng/g of total soluble protein. Bioassays revealed that the sugarcane lines were resistant to larval development and significantly reduced root damage caused by the larval of L. signata to 14.33. These results proved that this is an effective solution to improve sugarcane varieties, increase resistance to Scarab beetles in breeding and sugarcane production.
