摘要

Intramitochondrial supramolecular assembly can be a new therapeuticstrategy for treating cancer because mitochondria are the key for virtuallyall facets of the tumorigenesis. However, the in vivo applications ofmitochondria-targeting molecules are limited due to the positive charge andhydrophobicity that should be possessed by these molecules to penetratethe mitochondrial membrane, which may induce nonspecific serum proteininteractions and normal cell accumulation. Herein, a stimuli-responsivemitochondria-targeting molecule, Mito-SA that forms nano-sized micellesunder physiological conditions is presented. In the aggregated state, thesuccinic amide bonds in Mito-SA are cleaved in response to tumoral pH bystabilizing the transition state through the intermolecular interactions and themicelle disassembles into a mitochondria-targeting parent molecule, Mito-FF.The Mito-FF accumulate inside cancer mitochondria to induce cell death byintra-mitochondrial self-assembly into fiber structures. This tumoral stimuliresponsivedisassembly–assembly approach will provide insight to developmitochondria targeted supramolecular anticancer agent with high tumoralspecificity.
