The present research was envisaged to synthesize different grades of a novel, nontoxic, and tissue-compatible neem gum based polyvinyl acetate grafted neem gum amphiphilic graft copolymer (NG-g-PVAc) using the microwave-assisted free radical technique and it was further optimized for drug delivery applications. Grafting was confirmed for the optimized grade G7, using analytical techniques like, Fourier transform infrared spectroscopy (FTIR), ~(13)C NMR, thermogravimetric analysis (TGA), x-ray diffraction (XRD), and optical contact angle measurement (OCA). The critical micelle concentration (CMC) of G7 exhibited very low values displaying surfactant characteristics. Invitro release study revealed that about 95.05 of curcumin CUR) was released from the prepared tablet with G7 for 10 h, showing Korsmeyer-Peppas model indicating sustained release property. The permeability of curcumin was found to be 13.87 using the amphiphilic graft copolymer. The prepared amphiphilic graft copolymer displayed anti-microbial activity against Staphylococcus aureus indicating its application in wound healing. The optimized grade G7 enhanced fibroblast and collagen growth when inserted under the skin hypodermis in a mice model indicating the polymer can be used for tissue regeneration. Hemocompatibility studies suggested G7 to be biocompatible. Thus, the novel amphiphilic graft copolymer displays a promising future as a polymer scaffold for tissue regrowth.
