Unilateral acute renal failure was induced in the dog with an intra-arterial norepinephrine infusion (0.75 mu;gsol;kgsol;min) for 2 hours. Follow-up at 3ndash;7 days revealed oliguria associated with indirect indices of a reduction of cortical perfusioncolon; absence of a rapid component of133Xe washout and absent cortical nephrogram. In this study radioactive microspheres were used to assess the possibility of shunting and provide a more direct index of cortical perfusion. Acute renal failure was associated with a preferential reduction of cortical perfusion and no evidence of shunting was obtained. The response of the renal vasculature to acetylcholine suggested a major element of active vasoconstriction in the reduction of cortical perfusion. Because neither blockers of norepinephrine nor of angiotensin even in high dose delivered directly into the renal artery modified perfusion or function, it is unlikely that either represents the mediator of the vasoconstriction. Either the cortical vasoconstriction is due to an unknown, intrarenal mediator or represents a primary resetting of intrinsic arteriolar smooth muscle tone.
