Eight healthy volunteers received a 6-minute infusion of pantoprazole 80mg and placebo in a double-blind randomised crossover study. Cardiovascular function was investigated at rest and during exercise by measuring blood pressure, heart rate and ECG, and also during passive postural stress on a tilt table. Cardiac performance was assessed noninvasively by means of systolic time intervals (STIs). Serum concentrations of pantoprazole were measured by high-performance liquid chromatography. Following the 6-minute infusion, no clinically relevant changes in blood pressure, heart rate, ECG, STI or routine laboratory parameters were observed after placebo or pantoprazole. Both infusions were well tolerated. A minor influence of pantoprazole on orthostatic regulation of diastolic blood pressure could not be excluded during the tilt table test, but, if present, was judged to be of no clinical importance. It was concluded from the systolic time intervals that pantoprazole had no influence on cardiac contractility.
