Rapid production of type I interferon (IFN-α/β) is a characteristic response to many infections. IFN-α/β lays a key role in limiting early viral spread by inducing the expression of numerous genes that interfere with viral replication and by activating natural killer (NK) cells and macrophages to kill infected cells. Although most cell types are capable of expressing IFN-α/β, the capacity to secrete high amounts of type I IFN appears to be restricted to relatively rare cells. Several years ago, unusual cells were identified in human blood that secreted high amounts of IFN-α/β, expressed low amounts of cell surface CD4 and major histocompatibility complex (MHC) class II, but lacked hematopioetic-lineage markers. These cells were recently shown to correspond to a population of cells with plasmacytoid morphology that are present in the T cell areas of inflamed lymph nodes and are capable of differentiating into dendritic cells (DCs). As reported by Asselin-Paturel et al. in this issue of Nature Immunology and by Nakano et al. in the Journal of Experimental Medicine, it is now clear that an analogous plasmacytoid cell population that secretes high amounts of IFN-α/β also exists in the mouse.
