The formation of double strand breaks in the DNA (DSBs) of eukaryotic cells, either by physical and chemical agents, or through errors during DNA replication, initiates a highly coordinated network of reactions aiming at the restoration of genomic integrity through repair, or the demise of the cell through apoptosis or senescence. While frequently these reactions are conveniently separated in signaling, for example to checkpoints, and repair by homologous recombination or non-homologous end joining, it is becoming increasingly evident that signaling and repair are inseparable components of what is now referred as DNA damage response (DDR).
