Natural and synthetic marine adhesives were screened for use on enzyme-linkedimmunosorbent assay (ELISA) plates. The most effective in repetitive use was glucosamine polymer (GAP). Evaluation was based on percent of capture antibody bound to the immunosurface before and after chaotrope treatments. Storage of chaotrope-treated plates in phosphate buffered saline/1% BSA/O.1% thimerosal (DBT) +3% sucrose enhanced the immunoreactivity of the immobilized capture antibody and stabilized the plate for up to 1 month. Previous difficulties with sensitivity were overcome by obtaining the specific strain of Escherchia coli from which the anti-E. coli IgG2b and anti-E. coli IgM bound to aE. coli IgG2b, and the immunocomplex was developed with a secondary anti-Mouse IgG antibody labeled with alkaline phosphatase.
