Probing the Function of Engineered Proteins by Electrochemical Techniques

In this report, we propose an electrochemical approach to address this problem ina subclass of integral proteins that are photoactive and are either sensor proteins (which transduce light energy for the purpose of information processing) or transport proteins (which convert light energy into electrochemical energy). Our approach is based on the analysis of ultrafast light induced electric signals (known as displacement photocurrents) which reflect the biochemical events at each of the three domains. In addition to the spatial attributes, these photoelectric events also operate at a frequency domain beyond that of many conventional biochemical techniques. The electrochemical technique to be described permits detection of the electric signals associated with rapid charge movements in the three domains, and therefore provides a valuable tool to probe the protein functions, complementary to spectroscopic techniques.
