The insoluble protein-dye complex, Azocoll, can be used to measure proteolytic activity as a function of time. By using the aliquoted method discussed in this report, one 5.0 ml reaction mixture can give up to five points on a standard velocity curve. It has been shown that the ratio of solid to liquid in the reaction mixture can be kept constant to give accurate values when pipetting aliquots from a solid-liquid mixture. We have also shown that Azocoll is more sensitive to trypsin than previously reported and that the 'dye bleeding' in control samples can be kept to a manageable level. (Author)
