Unaudited draft pathology reports of 2-year inhalation studies in Syrian hamsters and Fischer 344 rats exposed nose-only to 30 mg/m3 of refractory ceramic fibers, 10 mg/m3 of asbestos fibers (positive controls), or clean air are presented. Animals were exposed for 6 hrs/day, 5 days/week. Individual animal data are presented. Pleural tumors were noted in 36/102 hamsters (35% incidence) exposed to the ceramic fibers. Of these, 32 tumors occurred between 66 and 89 weeks on the test. No lung cancers were observed in any group. Data from the 24 month sacrifice of rats are not available, but a review of some interim deaths revealed a malignant mesothelioma in one rat exposed to fiber. A multi-dose study was conducted in rats exposed for 26 weeks to refractory ceramic fiber concentrations of 25 fibers/cc, 75 fibers/cc, or 150 fibers/cc. No further information regarding protocol is provided. Unaudited draft pathology reports indicate that dose-related fully reversible cellular changes were seen in the lung. These were characterized as minimal fiber-containing microgranulomas at the bronchiolo- alveolar junction, fiber-containing macrophages, and free fibers in the alveoli of all rats. The changes were not fiber-specific, but were of the type expected from the presence of any foreign particulate in the lung.
