Studies in this laboratory indicate that members of the diverse spectrum of hepatic lesions appearing in rats given a single neonatal treatment with carcinogen followed by promotion with dietary phenobarbital are developmentally independent. The more recent experiments provide a more definitive evaluation of the developmental relationships among these lesions by subdividing the altered hepatocyte focus population into foci that were detectable solely by histochemical means and those that also showed morphological aberrations and comparing the developmental and phenotypic properties of the hist(+), morph-) foci, hist(+), morph(+) foci, and hepatic tumors in rats initiated once neonatally with different doses of diethylnitrosamine and promoted with dietary phenobarbital from weaning. Rats were killed at intervals, and their livers were examined for tumors and for the two classes of altered hepatocyte foci. Through application of serial frozen-sectioning techniques (2) and computer-assisted image analysis (3), foci containing between one and seven histochemical markers and/or exhibiting morphologic changes were identified and sized. The same complement of histochemical tests was applied to the hepatic tumors observed in this investigation. 3 refs. (ERA citation 13:019638)
