摘要

Fascioliasis is an important parasitic disease of grazing cattle and sheep. The experimental approach included the production of metacercariae of Fasciola hepatica from Lymnae tomentosa, infection of experimental animals, separation of fractions from whole Fasciola hepatica antigen by gel filtration on Sephadex G-100 and G-200, labelling of antigens with exp 125 I, and examination of experimentally infected sheep and other animals for antibodies by modified radioactive single radial diffusion, indirect radioimmunodiffusion, and radioactive antigen microprecipitin assay by means of direct radioimmunodiffusion and immunodiffusion using unlabelled antigen. Sufficient quantities of fluke for the production of antigen were obtained only from sheep. Infected rabbits, guinea pigs, and rats yielded relatively few flukes. Labelling of the antigenic fraction can be regarded as fairly successful but the specific radioactivity of the fractions needs to be increased. Results from antibodies in infected animals by indirect radioimmunodiffusion and modified radioactive single diffusion proved not fully specific. Radioactive antigen microprecipitin assay would appear to differentiate between negative and positive sera. (Atomindex citation 09:389323)
