A total of 2602 samples consisting of 511 sediment, 1753 various species of fish, 141 shellfish, 121 shrimp and 68 crab were collected from 69 fishing areas in the coastal water of North Java and East Sumatra for detecting the incidences of Cl. botulinum and V. parahaemolyticus. From these samples, 70 (2.69%) were positive for Cl. botulinum and 86 (3.3%) were V. parahaemolyticus positive. The positive samples of Cl. botulinum consisted of 15 type A, 13 type B, 20 type C, 19 type D and 7 type F. The incidences around the north coast of Java were mainly type C and D, whereas those around the east coast of Sumatra were mainly types A and B. Growth and toxin formation of Cl. botulinum type E (strain Iwanai) was completely inhibited in media containing 7.5% NaCl and above. Radiation doses of 200 krad and above together with NaCl content in the media of 5.0% can also inhibit growth and toxin formation of this type. Inoculated pack studies of Cl. botulinum type C in radurized salted mackerel showed that either a treatment with 300 krad or a NaCl content of 10% was able to delay toxin formation in the fish held at room temperature for 6 weeks. Irradiating fish containing 5% NaCl with 200 krad can also delay toxin formation in the fish held at room temperature for a minimum of 4 weeks. V. parahaemolyticus can easily be eliminated from radurized fish receiving a dose of 100 krad and above. The D-value of this microorganism in a fish media containing 3% Na Cl was approximately 20 krad. (Atomindex citation 08:332557)
