This paper begins with a description of lysosome theory;then it discusses the role of lysosomes in inflammation, inflammatory joint disease, and rheumatoid arthritis. It ends with comments on the lysosome-modifying actions of established drugs useful against rheumatoid arthritis and a discussion of approaches to future drug research in this field.nLysosomes are cytoplasmic granules, each bounded by a membrane;they contain many hydrolytic enzymes and other biologically active substances. They are in all cells, where they arise as primary lysosomes (see Fig. 1) from the golgi apparatus and endoplasmic reticulum. Later they discharge their contents directly out of the cell or into secondary lysosomes, and as they do this their membranes are fused with those of cytoplasm or phagosomes;other lysosonies and autophagic vacuoles become residual bodies and may eventually be expelled from the cell.
