The continued use of rotenone as a fish toxicant depends on the development of information requested by the U.S. Environmental Protection Agency for reregistration. To meet one of the requirements, the authors exposed Daphia magna to rotenone in toxicity tests. In exposure of 0.5 to 10 micrograms/L rotenone in a 48-h acute toxicity test, the EC50 was 3.7 micrograms/L; in exposures of 0.312 to 5.0 micrograms/L in a 21-day chronic toxicity test, the EC50 was 2.1 micrograms/L. The no-observed-effect concentration was 1.25 micrograms/L. In a flow-through toxicity test, the authors exposed eyed eggs and larvae of rainbow trout (Salmo gairdneri) for 32 days to crystalline technical grade rotenone to determine its effects on survival, hatching, and growth. Rotenone concentrations of 1.0 to 10.0 micrograms/L did not kill the eyed eggs which hatched normally after 5 or 6 days of exposure. Information required by the U.S. Environmental Protection Agency to support the continued use of rotenone as a fish toxicant includes data on toxicity to nontarget organisms. Information summarized here was developed in three mammalian safety studies of rotenone: chronic oral toxicity in rats, effects on reproduction in rats and subchronic oral toxicity in dogs.
