2'-Fluoro-5'-iodoarabinosyl uracil (FIAU) and 1-(2-'fluoro-2'-deoxy-Beta-D-arabinofuranosyl) thymine (FMAU) have been examined for antiviral activity against simian varicella virus infection in the African green monkey. Both FIAU and FMAU inhibited replication of the virus in vitro at a concentration of 0.1 micrograms/ml. Pharmacokinetic studies with FIAU showed plasma levels exceeding 0.1 micrograms/ml were attained for greater than three hours following a single oral dose of 5.0 mg/kg. Subsequent in vivo studies with FIAU at 1 mg/kg/day prevented rash and viremia while a dose of 0.2 mg/kg/day was without effect. When FMAU was compared at similar doses, 0.2 mg/kg/day prevented both rash and viremia. FIAU treatment initiated as late as 3, 5, or 7 days into the inoculation period inhibited the progression of simian varicella infection. FIAU at 30 mg/kg/day orally delayed as late as 7 days after virus inoculation reduced rash and hastened disquamation and caused a prompt inhibition of viremia. E-5-(2-Bromovinyl) arabinosyl uracil (BV-ara-U) also had a marked effect on simian varicella infection in the monkey. When decreasing doses of BV-ara-U were examined, an oral dose of 0.2 mg/kg/day begun 48 hours after virus inoculation inhibited the development of rash and reduced viremia.
