摘要

Adult rhesus monkeys were administered daily or weekly intravenous injections of the cancer chemotherapeutic agent F3TdR (NSC 75520) at dosages of 12.5, 25or 50mg/kg,or 200,400, 800or 1200mg/kg,respectively,for 90days. A comparison of toxic effects observed indicated that smaller daily dosages were better tolerated. Drug-related deaths occurred after a single injection of F3TdR at levels of 400-1200mg/kg. Severe liver involvement,confirmed histopathologically,was indicated clinically by elevated serum glutamic pyruvic transaminase (SGPT) values immediately after the initial treatment at dosages equal to or more than 200mg/kg and was accompanied by increased bilirubin levels at dosages equal to or more than 400mg/kg. These hepatic changes preceded erythropenia and/or leukopenia at the lower levels. Although clinical test values from animals receiving smaller daily doses of FeTdR also showed some liver involvement,the effect was less severe and occurred later. (Modified author abstract)
