The work during the last six months has been largely devoted to the investigation of methods to be used effectively disrupt adenovirus types 3, 4and 7with the emphasis on type 7. Fifty fold (50X) methanol concentrates were prepared with WI-38growth adenovirus type 7and disrupted at pH 10.5with tween 80for a variety of different time periods. The resulting materials were assayed for residual viable in HEK,WI-38and HeLa cell cultures and by complement fixation and animal potency. A disruption procedure was developed which recovered virtually all the antigen,as determined by animal potency,and was devoid of residual viable virus. This technology was subsequently employed in preparing a vaccine (type 7) as a possible candidate for clinical evaluation in man. Parainfluenza 1, 2and 3were propagated in embryonated eggs and amniotic fluids were collected for the preparation of future vaccines. Parainfluenza 1Challenge Pool (Lot 7286) has been completed and Parainfluenza 2Challenge Pool (Lot 1013) is near completion. (Author)
