Seven rhesus monkeys received up to 8 weekly treatments with daily oral doses of 100, 50, 25 or 12 mg/kg. The one monkey treated with 100 mg/kg became moribund on day 28. Toxicity included emesis, weight loss, dehydration, severe neutropenia, marked anemia and moderate elevation of BUN. Histopathological changes included severe depression of the bone marrow, severe lobar exudative pneumonia, marked focal necrosis of hepatocytes, and moderate toxic changes in renal, myocardial, intestinal and lymphoid tissues. The 6 monkeys treated with weekly doses of 50, 25, or 12.5mg/kg were kept for a post-treatment observation period of at least 38 days. All 6 monkeys survived. One of the 2 animals treated with 50 mg/kg/day was sacrificed in poor condition; all of the other animals were in good general condition at autopsy. Toxicity was generally dose related and included protracted anemia, reversible neutropenia and intermittent decreases in serum Na+, K+ and Cl-. all survivors exhibited intermittent proteinuria and urinary cast formation. Treatment with 12.5 mg/kg/day led to moderate, reversible body weight loss, anemia, leukopenia and electrolyte changes. All monkeys showed marked fatty changes of the hepatocytes, myofibers and moderate changes of the renal tubular epithelium. (Author)
