Provided herein is a method for developing a protein expression profile in a biological sample of formalin-immobilized culturing tissue obtained from a subject. The SRM/MRM assay provided herein detects proteins involved in cellular processes, including cell division, cell differentiation, cell growth inhibition, cellular metabolism, cell signaling, and tumor immune response/regulation directly in a subject's tumor tissue. And can be used to quantify. The SRM/MRM assay can provide a tumor tissue profile of the entire tissue microenvironment that indicates the optimal cancer therapy treatment for the subject, regardless of the cell origin of expression.
