The present invention relates to a sleep-promoting animal model through optogenetic stimulation of astrocytes in a ventrolateral preoptic nucleus (VLPO), and a sleep control agent screening method using the same. More specifically, an adenovirus including a photoreactive gene controlled by a GFAP promoter to control astrocytes in a living body is injected into a brain of a rat such that the photoreactive gene is expressed in the astrocytes. Moreover, an optical fiber is implanted to transfer an optical stimulus into the brain and blue light is irradiated through the installed optical fiber such that the astrocytes are stimulated. Through the present invention, it is confirmed that optical stimulation of the astrocytes in a VLPO promotes an increase of sleep and promotes activation of gliacytes and neurocytes. Accordingly, technology developed through the present invention can be useful for researching the control reaction of astrocytes in sleep promotion and provides possibility in an astrocyte-centered treatment strategy for controlling sleep and treating diseases related to sleep.
