Provided is a method for detecting an off-target effect of an adenine base editor (ABE) system based on whole-genome sequencing - EndoV-seq. The adenine base editor system consists of two components: a fusion gene of TadA:TadA*:Cas9 and a gRNA, wherein same can catalyze efficient replacement of an adenine (A) with a guanine (G) at a target site.
