The present invention belongs to the technical field of the preparation of precious metal nano materials and ratiometric fluorescent probes, and in particular relates to a method for preparing a melamine ratiometric fluorescent probe based on a composite of a DNA-stabilized silver nanocluster and rhodamine 6G. Electrostatic self-assembly is used to construct a silver nanocluster/rhodamine 6G composite; and a hydrogen bond is formed between melamine and thymine in DNA on the surface of the silver nanocluster, so that the rhodamine 6G is dissociated from the surface of the silver nanocluster, resulting in fluorescence resonance energy transfer being destroyed and silver nanocluster fluorescence being recovered. This process has a slight impact on rhodamine 6G fluorescence, so that same serves as a reference signal, and the silver nanocluster fluorescence serves as a response signal. A linear relationship between the ratio of the fluorescence emission peak intensity of the silver nanocluster to that of the rhodamine 6G and the molar concentration of melamine is fitted, thus constructing the melamine ratiometric fluorescent probe. The probe has a simple preparation process, is low in cost and high in product sensitivity, and can be developed as a novel melamine ratiometric fluorescent probe used for effectively detecting melamine in an actual sample.
