Immunotherapy, such as T cell therapy, and methods of treatment associated with administration of tryptophan metabolism and / or kinulenine pathway regulators are provided. In some embodiments, the methods comprise co-treatment comprising administration of tryptophan metabolism and / or kinulin pathway regulatory factors, such as engineered T cells, such as chimeric antigen receptor (CAR) -expressing cells, and inhibitors of enzymes . Also provided are engineered cells that have modified expression of molecules involved in the kinourenine pathway. Also provided are methods of preparing the engineered cells, cells, compositions, methods of administration, nucleic acids and kits. In some aspects, the methods and features of the cells increase or improve cellular activity, outcome, function, response, persistence, volume enhancement and / or proliferation for adoptive cell therapy.
