Leptospira bacterial agglutinating lectures of crotalaria spectabilis possible indication as markers in rapid diagnosis of leptospirosis. The present invention relates to the identification and purification for the first time of proteins capable of binding pathogenic leptospires in in vitro assays. Its binding activity on these bacteria occurs even when they are in low concentration, or when lectin has been diluted after protein precipitation. the proteins used in this invention were extracted from crotalaria spectabilis seed, which were precipitated in acetone (80%), lyophilized and isolated by filtration chromatography (sephadex a-100) followed by ion exchange chromatography (deae sephadex a-50). . Leptospirosis is an emerging zoonosis with worldwide distribution and is most commonly found in low-income populations in developing countries. Since leptospires also have sugars on their cell surface, our results indicate that the lectins of crotalaria spectabilis are able to recognize and bind to these bacterial surface sugars, serving as a possible marker in rapid diagnostic tests. The present invention should be supplemented for use as a diagnostic method for leptospirosis. Additional assays of bacterial specificity, protein toxicity and signal amplification will be required.
