The present invention relates to a nanoliposome carrier composition encapsulating Cas9 protein, a guide RNA suppressing the expression of KRAS gene, and a cationic polymer; or to a therapeutic agent for anticancer drug-resistant colorectal cancer induced by KRAS mutation comprising the same. Cetuximab, which is currently used as a drug for metastatic colorectal cancer, is only effective in patients with KRAS-normal colorectal cancer, and has a critical disadvantage that even in the patients with KRAS-normal colorectal cancer, when continuously treated with Cetuximab, 60-80% of these patients may develop into KRAS-mutant type. However, by using the nanoliposome composition of the present invention, mutations of KRAS, a colorectal cancer-causing gene, can be fundamentally prevented, and the treatment of KRAS-mutant metastatic colorectal cancer can be carried out very effectively.
