A method of screening a substance regulating MMP-2 secretion from a cell comprising using a cell transformed with a polynucleotide encoding a proMMP-2-luciferase fusion polypeptide. The screening method is useful for screening drugs such as MMP-2 secretagogues having an MMP-2 secretagogue activity which can be used as anticancer drugs, which may be MMP-2 secretion inhibitors or cancer metastasis inhibitors. The transformant in which a polynucleotide encoding the fusion protein used for the screening method is introduced, the screening kit comprising the transformant, etc. are also useful for screening other drugs. The proMMP-2-luciferase fusion protein and nucleic acids encoding said protein are claimed and the luciferase may be Gaussia luciferase.
