The present invention relates to a reaction mixture for the molecular detection of potato tuber spindle viroid by quantitative RT-PCR, characterized in that it contains 1 .mu.l of a sample of RNA tested, 12.5 .mu.l of 2x master mix containing a buffer mixture, DNA polymerase , deoxyribonucleotide, and magnesium chloride, 1.0 .mu.l of reverse transcriptase / RNase inhibitor mixture, 9.3 .mu.l of sterile distilled water, adding to the final volume of 25 .mu.l of 0.1 .mu.l each primer at 100 µM and specific nucleotide sequence QRT R_PSTVd: CTT TCT TCg ggT gTC CTT CC, QPCR R_PSTVd: CTC ggg AgC TTC AgT TgT TTC that amplify a 101 bp fragment and 1 µm probe 30 µM PSTVd and TgCgCTgTCgCTTCggCTACT specific nucleotide sequence, wherein the probe is 6-carboxyfluorescein fluorescent dye (FAM) and tetramethylcarboxyrhodine (TAMRA) sub is used as quencher. .n.
