A PCR method is carried out on DNA from a Japanese flounder using, as primers, oligonucleotides having the following nucleotide sequences: 5'-GTGGACATTT GTACTCCACA GACCA-3' and 5'-GTGAGCGGGT ACATGTGTGT GAG-3' for Poli185TUF or oligonucleotides having the following nucleotide sequences: 5'-CCTCACAAAG ATATTTGTAC AGGTGCA-3' and 5'-CATCTTTAGG TCACATTGTC ACTGCTG-3' for Poli109TUF. The products produced by the PCR method are subjected to gel electrophoresis. A Japanese flounder which has a band at 212 bp with respect to Poli185TUF is determined to be a male, while a Japanese flounder which has a band at 144 bp with respect to Poli109TUF is determined to be a male. This method enables to determine a genetic sex of a Japanese flounder without fault prior to the sex differentiation period of the Japanese flounder.
