Evaluating activity of an agent on epidermis renewal in vitro, comprises preparing equivalent of epidermis in the presence of the agent to be evaluated, where the epidermis equivalent is prepared from a population of keratinocytes
Evaluating activity of an agent on epidermis renewal in vitro, comprises preparing an equivalent of epidermis in the presence of the agent to be evaluated, where the epidermis equivalent is prepared from a population of keratinocytes in which the percentage of clonogenic cells with respect to total keratinocytes is modified by at least 10% with respect to the percentage of clonogenic cells of a standard population. Independent claims are included for: (1) a skin equivalent obtainable by the process, comprising the epidermis equivalent and at least one cosmetic or dermatological agent; and (2) screening cosmetic or dermatological active agents, comprising: (a) inoculating a dermis equivalent by a population of keratinocytes on a dermis equivalent in a culture medium, where the population of keratinocytes has a lower percentage of clonogenic cells of at least 10% less than the ratio of a standard population of keratinocytes; (b) contacting at least one cosmetic or dermatological active agent with keratinocytes or the culture medium; (c) leaving the epidermal equivalent for differentiating; (d) comparing the epidermal equivalent with control epidermal equivalent obtained in the same conditions but without the active agent to be evaluated; and (e) selecting the active agents for which the characteristics of proliferation and/or regeneration of the epidermis equivalent are increased with respect to cells of control epidermal equivalent.
