A method for producing a cell line expressing a stabilized functional human single chain antigen-recognizing TCR (scTCR), comprising a) providing a suitable host cell, b) providing a genetic construct of the human scTCR to be expressed, comprising the domains Vα / β-Li-Vβ / α-Cβ / α Vα-Li-Vβ-Cβ or Vβ-Li-Vα-Cα, where the linker Li represents any peptide sequence, and provide a genetic construct comprising the corresponding human hetero- / (Homo-) dimeric domain Cα / βCα or Cβ, by introducing amino acid exchanges that are alien to the human sequence into the domains Cβ / α and Cα / β, and. c) direct or indirect introduction of the genetic constructs into a recombinant cell via viral or non-viral gene transfer, and d) coexpression of the genetic constructs of the scTCR fragments by the cell.
