The present invention provides oligonucleotides, compositions that comprise them and methods that use oligonucleotides and compositions to stimulate cells that express the TLR7 and / or TLR8 receptor. Oligonucleotides to stimulate TLR7 comprise regions rich in uracil. Oligonucleotides to stimulate TLR8 comprise regions rich in guanine. The present methods and compositions are useful, inter alia, for treating or preventing conditions such as infectious disease and cancer.
