Conjugate, useful in detecting double stranded nucleic acids, comprises an uncharged intercalator, a spacer that is covalently bonded to intercalator, a first and second linker molecule and a detectable marker
Conjugate comprises: (a) at least one uncharged intercalator that specifically binds to a double stranded nucleic acid; (b) at least one spacer that is covalently bonded to the intercalator; (c) at least one first linker molecule that is covalently bonded to the spacer; (d) at least one second linker molecule that specifically binds to the first linker molecule; and (e) at least one detectable marker that is bonded to the second linker molecule and produces a detectable signal. The first linker molecule is biotin or biotin derivative. Conjugate comprises: (a) at least one uncharged intercalator that specifically binds to a double stranded nucleic acid; (b) at least one spacer that is covalently bonded to the intercalator; (c) at least one first linker molecule that is covalently bonded to the spacer; (d) at least one second linker molecule that specifically binds to the first linker molecule; and (e) at least one detectable marker that is bonded to the second linker molecule and produces a detectable signal. The first linker molecule is biotin or biotin derivative. The second linker molecule comprises avidin, streptavidin, neutravidin or a biotin-binding fragment of avidin, streptavidin or neutravidin. The marker is a biotinylated marker. An independent claim is also included for detecting a nucleic acid analyte, comprising: (i) hybridizing the target nucleic acid on a nucleic acid separator sequence to form a double stranded nucleic acid, (ii) incubating the double stranded nucleic acid with the conjugate to form a complex of the double stranded nucleic acid and the conjugate, and (iii) detecting the double stranded nucleic acid-conjugate complex.
