Identifying different species of genus Methylobacterium, comprises: (a) preparing a crude extract from suspension of methylobacteria; (b) amplifying 16S rRNA (rRNA) gene by PCR; (c) carrying out at least two restriction digests of the PCR products; (d) separating and analyzing the restriction fragments of two restriction digests; (e) detecting at least two restriction patterns of the parallelly arranged restriction digest; and (f) comparing the restriction patterns and DNA fragment length with the known restriction patterns of reference strains of Methylobacterium. Identifying different species of genus Methylobacterium, comprises: (a) preparing a crude extract from a suspension of methylobacteria; (b) amplifying 16S rRNA gene by PCR using the crude extract as DNA matrix and the oligonucleotides pA and pH as primers; (c) carrying out at least two restriction digests of the PCR products in parallel reaction batches; (d) separating and analyzing the restriction fragments of at least two restriction digests by gel electrophoresis; (e) detecting at least two restriction patterns of the parallelly arranged restriction digest; and (f) comparing the restriction patterns and the DNA fragment length with the known restriction patterns of reference strains of Methylobacteriumfor the identification of the methylobacteria strain in the crude extract.
