Structured illumination system for three-dimensional microscopy of sample, has beam generation unit formed of light source, modulators, diffraction network and filter, generating non-symmetrical coherent beams relative to axis of lens
The system (8) has a beam generation unit formed of a light source (9) e.g. laser, spatial light modulators (10), diffraction network and filter arranged for generating coherent light beams. A lens (12) including a rear focal plane (PFA) is arranged such that a sample (14) is positioned in a focusing plane (PMP) of the lens. Achromatic lens (19) focuses the light beams in the rear focal plane such that the beams interfere in a collimated manner in the focusing plane. The generation unit is arranged for generating two non-symmetrical coherent beams with respect to an optical axis of the lens. An independent claim is also included for a method for improving a three-dimensional microscopy of sample.
