ERR.gamma. is expressed in bone and cartilage in vivo and osteoblastic andchondrocytic cells in vitro. ERR.gamma.y is a transcriptionalactivator of an osteoblast-associated osteopontin (OPN) and chondrocyte-associated (Sox9) gene in osteoblasts and chondrocytesrespectively. Knockdown of ERR.gamma. expression by antisense oligonucleotidestrategies in osteoblastic cell cultures reducesalkaline phosphatase activity. Together these findings indicate thatERR.gamma. is expressed in and plays a functional role in bone andcartilage. The results also indicate that agonists and antagonists ofERR.gamma. may be useful as therapeutic agents in a wide variety ofdiseases affecting bones and joints.
