A nanoparticle-protein-hydrogel composite is provided to be used for a sustained-release drug delivery system and a sustained-release bone filler, which show the controlled initial release and have significantly increased sustained-releasing effect. A nanoparticle-protein-hydrogel composite comprises a polysaccharide-functionalized nanoparticle including a core consisting of a biodegradable polymer, an outside hydrogel membrane consisting of a biocompatible polymer emulsifying agent, and a polysaccharide physically fixed on the core and the hydrogel membrane; a protein specifically bound to the polysaccharide; and a hydrogel carrier which is a carrier of the nano-particle where the protein is specifically coupled and consists of a biocompatible polymer. A sustained release drug delivery system comprises the polysaccharide-functionalized nanoparticle; at least one protein drug which is specifically coupled to the polysaccharide and is selected from the group consisting of growth faction, chemokine, extracellular substrate protein and anti-thrombin III; and the hydrogel carrier. A sustained release growth factor delivery system comprises the polysaccharide-functionalized nanoparticle; an effective amount of growth factor including BMP or TGF-beta and at least one growth factor selected from the group consisting of VEGF, FGF, and PDGF; and the hydrogel carrier. A sustained release bone filler comprises the sustained release growth factor delivery system. A method for preparing a nanoparticle-protein-hydrogel composite comprises the steps of: (a) dispersing an organic solution by dissolving a biodegradable polymer in a non-cytotoxic organic solvent into an aqueous solution obtain by dissolving a polysaccharide and a biocompatible polymer emulsifying agent to prepare a polysaccharide-functionalized nanoparticle; (b) filling the polysaccharide-functionalized nanoparticle with a protein to prepare the protein filled polysaccharide-functionalized nanoparticle; (c) dispersing the protein filled polysaccharide-functionalized nanoparticle into a biocompatible polymer aqueous solution for preparing a hydrogel carrier; and (d) providing at least one crosslinking factor selected from the group consisting of a crosslinking agent, a crosslinking activating agent, and a physical crosslinking factor to the dispersion obtained from the step(c).
