An organic/inorganic composite electrolyte separator for a direct methanol fuel cell, and a method for preparing the electrolyte separator are provided to improve hydrogen ion conductivity and methanol barrier property. An organic/inorganic composite electrolyte separator is prepared by solution mixing 100 parts by weight of a poly(vinyl alcohol), polyacrylamide or polyethylene glycol base resin of a hydrophilic resin with 20-50 parts by weight of a hydrophobic sulfonated polysilsesquioxane nanoparticle having a cage structure and crosslinking it by using a crosslinking agent. Preferably the polyethylene glycol base resin has a number average molecular weight of 2,000-5,000; and the crosslinking agent is a multifunctional anhydride or a multifunctional isocyanate.
