A method for the in vitro production of over 50 kb long dna - constructs by directed, sequence-specific recombination, characterized in that it comprises the steps of:a) by two separate clone the dna molecules to be connected to each other in two bacterial plasmids in such a way that, in the resulting clones in each case at the ends to be connected to the dna molecules a specific dna - recognition sequence which is connected as a specific site;b) propagating of the plasmids which are obtained in step a);c) isolating the plasmids from the bacterial chromosome in agarose gel by cleavage of the bacterial chromosome position with a restriction nuclease, which is not the plasmids and subsequent separation of the split, bacterial chromosome; andd) in vitro connection of the two dna - molecules from the two plasmids in the presence of a temporarily molten agarose - gel matrix by a directed at the specific site of recombination.
