A process for the production of mycophenolic acid by culturing Penicillium brevi-compactum under weak acid condition is provided to improve productivity of mycophenolic acid in industrial scale by optimizing cultivation conditions, especially hydrogen concentration condition. The mycophenolic acid represented by the formula(3), wherein R1 is methyl or hydroxymethyl and R2 is hydroxy group or amino group, and its derivative are produced by culturing Penicillium brevi-compactum Dierckx ATCC 46514 in a medium at pH 4.5-6, preferably pH 5.4-5.6, wherein the medium contains a nitrogen source selected from an inorganic nitrogen source such as NH4NO3, KNO3, (NH4)2SO4, NH4Cl and urea, or an organic nitrogen source such as Nz-amine, bean proteins, concentrated and fermented corn steep liquor, cottonseed meal, soybean cake, skim milk, peptone, beef extract, yeast extract, CGM(corn gluten meal), casamino acid and casein; and the mycophenolic acid is esterified into mycophenolate mofetil(MPM)(2-morpholinoethyl (E)-6-(1,3-dihydro-4-hydroxy-6-methoxy-7-methyl-3-oxo-5-isobenzofranyl) -4-methyl-4-hexenoic acid) for oral administration.
