IN-VITRO MODEL OF BLOOD-BRAIN BARRIER, IN-VITRO MODEL OF DISEASED BLOOD-BRAIN BARRIER, AND DRUG SCREENING METHOD, ANALYSIS METHOD FOR FUNCTIONS OF DISEASED BLOOD-BRAIN BARRIER, AND ANALYSIS METHOD FOR PATHOGENESIS USING THE SAME
It is intended to provide a screening system for a drug which passes the blood-brain barrier and acts on the center, a drug which acts on the blood-brainbarrier per se, or a drug which is not expected to act in the center butmigrates into the brain. Further, another object of the invention is torealize a pathogenesis analysis research under pathologic conditions or thescreening by imposing various pathologic environments on the screening system.A blood-brain barrier in vitro model is prepared by using a three-dimensionalculture device comprising a culture medium, a plate for holding the culturemedium and a filter soaked in the culture medium and placed such that it doesnot come into contact with the bottom surface of the plate, wherein the filterhas many pores with a diameter of from 0.35 to 0.45 µm, and inoculating aprimary cultured brain capillary endothelial cell on the upper surface of thefilter, inoculating a primary cultured brain pericyte on the lower surface ofthe filter, inoculating a primary cultured astrocyte on the inner surface ofthe plate and co-culturing them with a standard culture medium.
